چکیده
Osteoarthritis (OA) is the most common joint disease. In last years, research community has focused on understanding molecular mechanisms that led to pathogenesis of disease, trying identify different and clinical phenotypes along with discovery new therapeutic opportunities. Different types cell-to-cell communication have been proposed contribute OA progression, including mediated by connexin43 (Cx43) channels or small extracellular vesicles. Furthermore, changes in chondrocyte phenotype such as cellular senescence contributors changing paradigm The use drugs able restore phenotype, reduce senescence-associated secretory components, modulate ion channel activity Cx43 appears be promising strategies for OA. this review, we aim summarize current knowledge related aging tissue damage opportunities currently available.

Structure of a Potential Therapeutic Antibody Bound to Interleukin-16 (IL-16): MECHANISTIC INSIGHTS AND NEW THERAPEUTIC OPPORTUNITIES.
Interleukin-16 (IL-16) is reported to be a chemoattractant cytokine and modulator of T-cell activation, and has been proposed as a ligand for the co-receptor CD4. The secreted active form of IL-16 has been detected at sites of TH1-mediated inflammation, such as those seen in autoimmune diseases, ischemic reperfusion injury (IRI), and tissue transplant rejection. Neutralization of IL-16 recruitm...
متن کاملCellular senescence and the senescent secretory phenotype: therapeutic opportunities.
Aging is the largest risk factor for most chronic diseases, which account for the majority of morbidity and health care expenditures in developed nations. New findings suggest that aging is a modifiable risk factor, and it may be feasible to delay age-related diseases as a group by modulating fundamental aging mechanisms. One such mechanism is cellular senescence, which can cause chronic inflam...
متن کاملNew immunosuppressive drugs: mechanistic insights and potential therapeutic advances.
Together with CsA, the new macrolide immunosuppressants FK506 and rapamycin have proved to be valuable tools in providing new information about key molecular events that underlie lymphocyte activation and degranulation. Studies of their mechanisms of action have pinpointed the phosphatase calcineurin and protein kinases as important signaling mediators in T-cell activation. Other new immunosupp...
متن کاملMechanistic and therapeutic insights gained from studying rare skeletal diseases.
Rare bone diseases account for 5% of all birth defects and can cause significant morbidity throughout patients' lives. Significant progress is being made to elucidate the pathophysiological mechanisms underlying these diseases. This paper summarizes presentation highlights of a workshop on Rare Skeletal Diseases convened to explore how the study of rare diseases has influenced the field's under...
